The Sunday night headline show will be the band's first UK festival since Knebworth in 1976, when they were joined by the likes of Lynyrd Skynyrd and 10cc.
The 27-date tour, which begins in Belgium on June 5, is the latest leg of the band's long-running "A Bigger Bang" tour, which began in 2005.
Jagger, Keith Richards, Ronnie Wood and Charlie Watts will play an additional UK date at the 02 Arena, formerly the Millennium Dome, on August 21. The band last performed on the Isle of Wight at a town hall in 1964.
A concert at Slane Castle in Dublin has been scheduled for August 18, while other gig locations include St Petersburg and shows in Serbia and the Ukraine for the first time. The tour will end on August 21 with a show at London's Millennium Dome, rebranded the O2 Arena.
Jagger, who works out and performs breathing exercises before taking to the stage, said: "I never get tired."
"I have a very definite countdown before the show where I have time to relax, get dressed, warm up."
He joked of Richards, also 63, who fell out of tree while on holiday in Fiji: "I'll nail him to the nearest room. I don't think he will be climbing any palm trees."
The Stones were forced to cancel a string of European dates on their tour last year. In April 2006, guitarist Keith Richards suffered a head injury, reportedly from falling out of a palm tree while vacationing in Fiji. Singer Mick Jagger contracted laryngitis in August.
The Stones had the top-grossing tour of 2006, amassing US$138.5 million (€104 million) in gross receipts, according to music trade magazine Pollstar.
Former Spice Girl Melanie C has also announced she will be performing on the Sunday, alongside the likes of James Morrison and Keane, while other new additions over the weekend include Mick Jones' band Carbon Silicon and Country Joe Macdonald.
The sold out festival takes place at Seahorse Park, near Newport on the Isle Of Wight, from 8-10 June.
Those already confirmed include headliners Muse and Snow Patrol, plus Paolo Nutini, Amy Winehouse, Groove Armada and The Feeling.
